چکیده

Since its original description, the endoplasmic reticulum (ER) has been considered to be a system of flattened cisternae interconnected with canalicular bridges or tubules (1) . This three-dimensional model of the ER was inferred mainly from images obtained on spread cells in toto where the ER appears as a continuous branching network . Subsequently, Palade (2) reported that the ER cisternae were not only branched and connected with bridges but also fenestrated . However, since thin sections for electron microscopy give only a limited sampling of an entire ER sheet, the number and the distribution of the fenestrae could not be accurately assessed . By exposing large areas of cellular membranes, the technique of freeze etching retrieves the threedimensional image lost by sectioning, and this communication will present data from freezeetched preparations showing clear images of fenestrae in the ER cisternae of the exocrine pancreatic cells of several species .

Gating Behavior of Endoplasmic Reticulum Potassium Channels of Rat Hepatocytes in Diabetes

Background: Defects in endoplasmic reticulum homeostasis are common occurrences in different diseases, such as diabetes, in which the function of endoplasmic reticulum is disrupted. It is now well established that ion channels of endoplasmic reticulum membrane have a critical role in endoplasmic reticulum luminal homeostasis. Our previous studies showed the presence of an ATP-sensitive cationic...

Ultrastructure of pancreatic exocrine cells of the rat during starvation.

Ultrastructural changes of the pancreatic exocrine cells after 3, 7, 14, 21, 28, 35 and 42 days of starvation were observed in male rats aged from 16 to 18 months weighing between 600 and 700 grams. The number of zymogen granules after starvation decreased to less than about 70 per cent of that of the control. Changes in the rough endoplasmic reticulum were hardly seen up to 14 days of starvati...

تمایز بن‌یاخته‌های‌ جنینی‌ انسان‌ به‌ سلولهای‌ مولد انسولین‌

Introduction: Type I diabetes mellitus is caused by autoimmune destruction of the insulin-producing β-cells. A new potential method for curing the disease is transplantation of differentiated insulin- secreting cells from human embryonic stem cells. Methods: Human embryonic stem cell lines (Royan H1) were used to produce embryoid bodies. Differentiation carried out by growth factor-mediated se...

Fine Structure of Differentiating Mouse Pancreatic Exocrine Cells in Transfilter Culture

Fine structural observations have been made in the 11-day embryonic mouse of exocrine cells in pancreatic epithelium developing in tissue culture transfilter from salivary gland mesenchyme of the 13-day embryonic mouse. After 2 days in culture, the exocrine cells show increased cytoplasmic density, abundant ribosomes in aggregate or "rosette" form, and expanded profiles of rough-surfaced endopl...

Apoptosis of mouse pancreatic acinar cells after duct ligation.

It has been established that pancreatic exocrine acinar cells disappear after pancreatic duct obstruction. This study aimed to examine the relationship between the disappearance of the acinar cells and apoptosis after pancreatic duct ligation of the splenic lobe in dd-mice, six weeks of age. In some mice, the ligature was removed after two or three days. In addition to general light and electro...
